No boss battles besides dropping Bowser in lava would be a missed opportunity, but it wouldn't hurt the game too much.
I've seen a level start with a "mini-boss fight" with Bowser Jr. in the Clown Car throwing shells. You had to beat him to use the Clown Car to move on. It was in the Mario World style and I liked how Jr. looked in that style.

@NintyMan - Yeah, it's the closest thing to a mini-boss though; I mean even Bowser is treated as just a stronger enemy so actual boss fights can't happen. Which is a shame but I guess doing that would mean adding an event-list feature of sorts and that might be over complicated compared to the current simplicity of it all.
I love what I've seen so far but the things that Super Mario Maker lacks for me are checkpoints and Star Coin-type collectables. I haven't seen them anywhere in the game.

I expect a direct closer to the time showing off more/unseen features from the game before it's released, with that being said I doubt it would include Boss fights altough I'd love to see Birdo there even as a mini-boss.
And regarding traditional boss levels, I guess that goes beyond what Mario Maker is designed to be. A good boss level needs unique AI, cut scenes, difficulty that increases in steps, dialogue, a cinematic end, etc.
Compare this to what we saw last year, so there's a chance that slopes, maybe even Koopalings and those rhino things, and caterpillar blocks and coin trails could show up in the final version.
Are you sure those things aren't already in the game. If coin trails is what I think it means, it's just coins attached to a trail. (There are many E3 examples of objects attached to trails.) And koopalings could be a skin of bowser jr (the skin/appearance of an object is changed by shaking the object).
I don't know their official names, the caterpillar blocks are those set of blocks that move a set path. The coin trail thing spawns from a ? block, it's basically a coin that travels a path that you control via the D-Pad. It also leaves a trail of coins.

Somewhat, but I think those just go in a straight line (they were originally used to float in the lava), the blocks I'm talking about can change directions and move in a complex path and all that.

With a Luigi amiibo, Luigi can be put in an item block. I've also seen footage of Luigis being shot through a canon as projectiles.
Don't know if Luigi will have any other jumping mechanics than Mario, though.
From the Treehouse stream: amiibo-supported characters are available in original 8-bit Mario mode only. Only changes appearance and sound effects, not movement.
From the Treehouse stream, it looked like the menu for unlocked amiibo costumes is massive. I think I might have even seen the number of spaces go up to 80!

They can't include every enemy, power-up or object, so expect a lot of complaining.
Why not?
Because there's a lot of situational stuff. Like a lot of levels from the NSMB series, they had a specific mechanic tied to each level, I don't think they're going to include everything. I'm sure they want to keep it simple, it shouldn't become a complicated level editor.
