Well Left 4 Dead is a pretty easy 'pick up and play' game. If they can make a quick matchmaking system for online matches I can see it work out great. The only problem would be the controls.

Ultimately, it would be up to Valve to develop something like this; knowing Valve, even if they started working on it right now, the earliest we'd probably see it would be 2014.
I don't see it happening. To my knowledge, Valve hasn't shown any support or interest in supporting Nintendo consoles, and that's not even considering the fact that they still have yet to make a portable game. Lastly, judging by how long their average development time is, it would probably take until the NEXT system after 3ds to make it happen.
Let's make [insert game name here that has no relevance to Nintendo]
QUEEN OF SASS
It's like, I just love a cowboy
You know
I'm just like, I just, I know, it's bad
But I'm just like
Can I just like, hang off the back of your horse
And can you go a little faster?!

@irken004 & @edhe : There's a perfectly good touch screen, which would be better to use and far more accurate than dual-analog sticks. Seriously, #$&%! dual-analog sticks for 1 st/3 rd person shooters.
OT: I'm going to say it's not happening. While it would be awesome, most of us have absolutely no idea if the 3DS can even run the Source engine, much less run it at acceptable speeds. And besides, a game like Left 4 Dead requires lightning reflexes, something that the 3DS' tiny screen can't quite encourage.

I'm not gonna say no, but I'm gonna say very unlikely. Valve has mostly been on the PC and Xbox 360. Barely now with Portal 2 did they move to the PS3 as well, and they did it late. But I wouldn't buy it. Small screen, graphic downgrade. There would be too much stuff on-screen. Unless they made it top-down, Pixelforce style...

The touchscreen is hardly suitable for first person shooters. Would you tap the screen to shoot?
Alternately, the analog could be used for movement, the face buttons for shooting and other functions, and the gyroscope (à la LOZ Ocarina of Time 3D) could be used for the camera.
You'd probably have to sit in a swivel chair to play it though.
