I think it'll either be streetpass content (Super Mario 3D Land: Special new Mystery Box; Mario Kart 7: Time Trial records/Mii characters from Nintendo; Kid Icarus: Uprising: Weapon Gems from Nintendo), or just minor updates (possibly in anticipation of MiiVerse) as @Morphtorok said. Or a bit of both. Anything beyond that... probably not.
Obviously if the Wii U is struggling now to beat the PS3/360 it will find a lot tougher competing with PS4/ONE. Anyway that's not what this topic is about.
@MK7 Why have an update for Street pass when you live in a place where you are lucky to get 2 a year. Like I said before a 'Continental' Mode online would prove better use.

There could be more to that than streetpass though!
1. Your point is not clear to me. Yeah this update could be to do with Nintendo Network, but it is still possible that there could be tweaks to the game itself

If it's through nintendo zone, then it will be just street pass content.... Because making an update and having it available only in certain places would be unfair and the update would need to be insignificantly small to be transmitted through those means and in these circumstances.

You see, as Mr GiokuShy tries to say, NintendoZone's streetpass relay works as if you walked next to another person with a 3DS... It exchanges street pass information in a quick manner.
I think the update they mentioned was supposed to be for the streetpass relay itself for it to support information from those games, not for the games themselves.
That means that after the update you will be getting not only streetpass hits for your mii plaza every time you pass through a nintendo zone.... You'll get hits for mario kart 7, mario 3d land and kid icarus too.
Nintendo Network is the 3DS and Wii U's online service. Nintendo Zone is a simple hotspot at participating locations (like McDonalds or Best Buy) that allows you to access specific Nintendo related materials (Coupons, advertizements, ect) and distributes streetpass hits to systems that enter its proximity.
@Morphtorok: You could already get streetpasses for those through streetpass relay... I think they're just distributing special streetpasses for those games.
@Ecto-1: And you can also access Nintendo Network from a Nintendo Zone. It's like free internet specifically for your 3DS.
