What it needs is higher difficulty, more levels, more exploration levels, unique musics, much harder bosses, and be able to use Yoshi in any level and even the in overworld (Boss stages are exceptions).
It must have Mario travelling to strange worlds in outer space, through fantastic means. Gravity must also be a big part of the game. Oh, and there should be a HUB world, too, which lets you play levels in any order.
Bowser in SMB1 was challenging... but mostly because I never got to him with Super Mario (in World 8, that is) and had to be extra careful. Lost Levels was even worse as I think you had to fight two, if I recall correctly.

Okay, I'll give you those, although I think in SMB1 you could run right under him a lot of the time.
V8, how on earth can you say you want the game to be harder and then suggest allowing Mario to take over twice as many hits? That makes no sense. It's not 3 strikes and you're out. It's one strike, find a coin, get another strike, find another coin, ad infinitum.
you've got a point there. Hmm...I should really rethink that statement. I probably meant to say that I want the game to be harder because you can take more hits (I'm pretty sure that doesn't make sense either, but I've been up for over 16 hours playing video games or being on the interwebz, so I could care much less at this point ).

So what you're saying is that it should be harder by making it easier to die. Are you also hoping the objective of the game is changed so that the goal is to die? That's a little dark for a Mario game.
Also, does it really get any better than an inflatable Yoshi?

My expectations are low because I know that way it will be great regardless. All I want is another 120 Super Mario Galaxy stars because that alone would be awesome. It doesn't need to be harder, it doesn't need to have different controls because the first one was fine. Just 120 more of the same quality of stars thanks.
As for things that I think would be nice little extras. I don't like water levels, they annoy the hell out of me although not so much after I worked out how to control it properly. Another thing, in the first one there were only a handful of levels that had 2D sections with the crazy gravity mechanics. I know New SMB Wii has covered that but I'd really liked those parts and would like to see them again. Oh, and more ice mario wall jump waterfalls and less spring mario.
