Mine is The World Ends with You. The story, complete with its memorable characters, teaches a great lesson about the importance of others. (If you didn't feel for Neku, then shame on you .) Not to mention, the gameplay is wild and exciting and makes great use of the Nintendo DS. The soundtrack is also amazing and the graphics are hip and cool. This is basically a graphic novel you can play. My only gripe is that it has only one save file. I love this game.

I got the Soundtrack on my iPod. Great tunes, every one (well, most of them).
Pretty tough choose but I'm gonna say Mario Kart DS. The best game in a legendary racing series with a new mission mode, twice as many tracks as usual (all being from the previous games) and basically the launch title for Nintendo Wi-fi? Works for me.
The other ones that are close are Kirby Canvas Curse, Advance Wars DS, Chrono Trigger and the 3rd Phoenix Wright game.

Henry Hatsworth Pokemon Soulsilver Zelda DS (both are virtually the same which isn't a bad thing at all)
Henry Hatsworth is this ultimate surprise. Fantastic graphics, and intense gameplay. Really hard and thus super rewarding when you finally make some progress!
Hotel Dusk: Room 215. It is most likely due to my love of reading, but I am obsessed with this game! The story is fantastic, the characters are intriguing, and the puzzles were fun. runners up would be any game from the Professor Layton series, Trace Memory, or Speed Racer.
