I think Nintendo is going to have a weak show this year. Their big titles are hitting this spring/summer for the most part, so I don't think we'll get much from them.
Microsoft and Sony will no doubt trot out their own heavy hitters, though.

Well, we don't know much about the 2nd half of the year, so I'm not so sure about that.
In my dream world it will be all about Star Trek and Nintendo will announce they're the new franchise licenseholders and we'll have a load of new Star Trek IPs - wouldn't that be great?
Definitely Zelda Wii. I'd say Last Story is a good bet, too. Ninty will most likely show off a new Wii xxxx game that uses the Vitality Sensor. Pikmin 3 seems very likely, as does Kirby Wii. I'm hoping Retro Studios' project will be shown and turn out to be Star Fox. Oh, and Intelligent Systems has got to be working on something. Paper Mario 4, anyone?
Can't forget the DS, either. Golden Sun and Dragon Quest IX will probably be the big DS showings. I'm hoping they'll announce a new DS(2?) Mario game too.
As far as hardware goes, I wouldn't be surprised at all to see the DS2. And of course the Vitality Sensor's going to be getting some screen-time too.
The conventional wisdom is be that Nintendo wouldn't be chucking out so many first party games in the next six months if they didn't have something big that they could count on for Christmas, so we're sure to see something big at E3. My money would be on a new Zelda or whatever Retro Studios have been working on for the past three years. I'd also be surprised if we didn't see some new Wii-branded game be it more sports, fitness or something else entirely.
In terms of hardware, I'd be surprised if we see a totally new console, although a new iteration on the DS and/or some extra peripherals might make an appearance.

I'm hoping! Perhaps they could announce a new TV series to be launched alongside the new game(s).
Going from the information we know, the new Zelda and Pikmin-games should be at E3 and I am looking forward to both. But Retro Studios mystery game is certainly the most intriguing. Could it be Kid Icarus? There were rumors about The Conduit 2 a few days ago as well. I wouldn't say no to that. Could be great now that they have learned from the few mistakes the original had. Also, I hope to see more from Dragon Quest X, Xenoblade and The Last Story. Love RPG's and these should be awesome.
And that is just for starters. What about the Vitality Sensor and it's games? My hope is a new Eternal Darkness. I have a feeling this E3 will be great.
@Imerion: Don't you mean Dragon Quest X ? I'm hoping we'll get to see it too. I've speculated that that's what Level-5's mystery project is, since they dev'd DQ8 for the PS2. :3
Im thinking Zelda Wii, Pikmin 3, and The Last Story will be talked about with trailers and game demoes. And some games for the Wii Vitality Sensor. And Probably whatever Retro Studios is working on and (hopefully) Project Sora. Like Sean Aaron said, they wouldn't Give us SMG2, S&P2, and Other M the first half of the year and leave us nothing for the rest of the year.
I'd really love to see more third-party games for the Wii. And see them getting a lot more promotion than they currently tend to. I'm not a dedicated Mario/Zelda/Metroid fan (though I do like them) so I'd like to see more third-party games for the console which will be worth getting and will sell well.
