I think it's be really cool if they made a replica to Samus's arm cannon. You'd hold it up like a wristwatch to check your health, your map, your missile count, change into morph ball, or scan everything. Your arm would get tired really fast hold it up all the time though, so I think it'd be better if the gun only showed up on screen when you're aiming, otherwise it'd just be resting in you lap. Also, a nunchuck would have to be used for movement, otherwise that would be really uncomfortable too.
For Wii U, it wouldn't be the worst thing if they stuck with the format of Prime, especially if they used the map on the pad screen or simply had the whole gameplay there, which would allow them to expand on the multiplayer from Echoes/Hunters. Or they could improve on the gameplay of Other M.
I definitely hope that a Wii U Metroid just doesn't use the Gamepad for scanning, and I'm with the user above, I'm ready for a Metroid Prime Hunters sequel or continuation. I know a lot of fans want Metroid 5, but it's had two chances to appear over nearly 11 years. They just don't know what to do there.

A sequel to Metroid Prime Hunters? I have never played the first one, but a second one will be AWESOME. If Retro said they want to expand the hunter universe, that could lead to the potential of a new Hunters game. I would like that too.
Just a small thought. I'm trying to think of a good competitive multiplayer shooter that's still fun with only local multiplayer. Having a 1 on 1 gun duel is kind of boring in most multiplayer shooters nowadays, and I can't think of a real way to fix this other than changing the goal from killing each other to killing something else, which would essentially make the game co-op. The main problem, in my opinion, is that when you only have one or two other players, there's just not a lot in terms of things to shoot at. In a 32-64 player online match though, the chaos is enough that you don't have to hunt for each other and there's always an enemy just around the corner. How do you solve this for a game that may wish to be local multiplayer only, to give everyone a lot of targets to shoot at without making the game co-op or adding in more players?
That said, a Left 4 Dead like co-op mode or a Halo Firefight like mode would be awesome.
