1. King Dedede (Kirby Series) He's undeniably cute, but has a giant hammer. And his true motives are usually hidden. 2. GLaDOS (Portal Series) She's kinda what made the portal games as good as they are. 3. Rosalina (SMG series) I would consider her the only Mario princess that doesn't annoy me.

These, and I would add:
4. Nathan Drake (Uncharted series) He's more believable than Laura Croft, and is what I consider the first real "Indiana Jones" of gaming. And Nolan North is awesome.
5. Toon Link (Zelda: The Wind Waker) His cat-like eyes and childish demeanor give him a better personality than the boring, drab Adult Link.
6. Master Chief (Halo series) He's the ultimate hero, badass, soldier, you name it. He's been to hell and back, and lived to tell the tale.
7. Samus Aran (before Metroid Other M) See Master Chief.
8. Kratos (God of War series) He's a force to be reckoned with. A whirlwind of anger, fury, and vengeance. He has the power of the Greek Gods at his disposal, and nothing can stop him.
9. Solid Snake (Metal Gear Solid series) He makes smoking look cool, and he gets the job done.
10. Sonic the Hedgehog He's fast, cool, and loves chili dogs. And Roger Craig Smith is his best voice actor yet.
11. Dr. Eggman (Sonic series) He may be a bumbling genius, but he's no match for Sonic's blazing speed.

3. Ezo Auditore - Assassin's Creed II One of the few game characters that you watch grow from a brash young man to a wiser older master. His character development was one of the most interesting in my opinion.
4. Gordon Freeman - Half-Life: You ARE the Freeman. His thoughts are whatever your thoughts are. No supernatural powers, just a man with a PHD in Theoretical Physics from MIT. Yet he has shown the world and aliens that he can kick major ass.
5. GLaDOS - Portal/Portal 2 - She made the Portal series the awesome games they were.
6. Godot - Phoenix Wright: Trials and Tribulations One of the most memorable prosectuors in the series. He sports an awesome visor and makes drinking coffee so badass.
Dr. Wily - He's more threatening than that bumbling Dr. Eggman / Robotnik.
Mega Man - He's the best thing Capcom has created (next to Street Fighter and their "VS Capcom" games).
Mario - Nintendo's mascot and all-around good guy. I haven't played a bad Mario platformer yet, all of his adventures have been fun in their own way.
Link - I LOVE THE ZELDA SERIES!!!
Sonic - He's fast and cocky, plus he loves chili dogs.
GLaDOS - Her sarcasm and ridicule made completing each puzzle more entertaining. Without her "guiding" you, Portal would have been incredibly boring.
As for modern characters... I don't really know of any standouts. All the soldiers in Killzone, Call of Duty, Resistance, etc. are all cookie cutter generic characters with dull personalities. Not my thing. I play video games to escape from reality, and cartoony characters are just the ticket I need.
On the other hand, I love well thought-out characters like Nathan Drake and Snake. They have established personalities and motives. Nathan loves exploring for treasure and learning about his family history. As for Snake, I haven't played much of the Metal Gear Solid games, but I enjoy the concept of the series.
1. Kirby - I don't know why... I just fell in love with the character when I first saw him and everytime I saw a Kirby game, I'd want to buy them immediately even if I couldn't. I even enjoyed Kirby's AirRide- though, not just because it is a Kirby game
Plus, absorbing enemies powers and using them against them is fun. Favorite has to be the Bomb Ability and UFO.
2. Ness - Although Ness doesn't speak, I like his simple character design and reminds me so much of "The Peanuts". Earthbound was also my first RPG. Earthbound is the only RPG I played through more than once, twice and thrice...
3. Clank - Love this little robot. Full of character. I love how he developed more and more after sub-sequential releases of Ratchet and Clank. As far as I can remember, Ratchet and Clank didn't really get along at first and now, they are best buddies. As should be.
4. Wario - My favorite character from Mario's universe. He's greedy, narcissistic and just plain funny. He's so much better than that pansy plumber Mario :3
5. Ulala - Probably my most favorite female protagonist in the video game world. She's charming, pretty and most importantly, groovy. She puts most reporters in the real world to shame. And I'm waiting for Space Channel 5 Part 3, Sega >:[
EDIT: I have so many characters going through my mind, I missed two more!
Viewtiful Joe and Silvia. Joe's humor, although cheesy, I find quite funny. And Silvia.. Well, she's feisty and she sports a nice outfit.. Why couldn't she be in UMvC3 as a playable fighter..?

1>Ramona Flowers (Scott Pilgrim Vs the Universe) She's a flawed, adored and fortunately not-often pawed fem fatale... who can spawn random items such as ninja swords.
2>Kamille Bidan (Gundam Dynasty Warriors 1,2,3) Kamilles a man's name, and he's a man.
3> Optimus Prime (Various) For keeping on trying despite yet finding a good game to star in. Nice try with War on Cybertron... nearly there!
4> Optimus Prime (Various) Surely the amount of times he's died and come back was the inspiration behind the 'life' system???
5> Ash Ketchum (None of the Pokémon games) The most famous name to NEVER star in the game he's famous for - Ash has Red & Blue's photos on his dartboard!

Samus Aran is my favorite video game character of all time, and I don't see that ever changing. I doubt I need to go into detail as to what makes her so likeable...
