I'm not sure if I should be voting considering I contributed an entry...
Still, regardless of that, I really want to vote for number 2: Oak's Lab. Late's pixelwork is amazing and the puzzle took me quite a while to solve. It was nice because it wasn't overly frustrating yet at the same time it was very satisfying once I figured it out.
I can't even lie. I had a lot of fun making my own puzzle and playing it, but my puzzle doesn't have that same level of difficult satisfaction.
It would be nice to have more people join and vote although this time there seem to be more voters than before which is really good thing. Winning will be harder with more people with different tastes and different reasons to vote. I myself am always looking for challenge as I'm really big fan of puzzle games but it's always good when the stage is also looking good. I'm also hoping to have some kind of competition where there's no way to use any existing pixel art. It would bring a nice fresh twist to this competition.

I'm a big fan of puzzles based on pixel art, but at the same time I'd be interested to see a competition like that. Even if I couldn't figure out a puzzle to make myself, it would be interesting to follow and vote on.
Also, slightly off topic, but considering how much you seem to really like difficult puzzles, I'm kind of curious how far you got in the main game? I've been stuck on the final set in the Bonus Pushmos for quite a while now to the point where I've practically given up on them and it's almost impossible to find any information regarding those bonus pushmos at all.
It's a tough call, but I'll put my vote in for #2.
Regarding this talk of original puzzles - looking back at the Warp Ladder Mania theme... unless I'm mistaken, none of those entries are based on existing pixel art. I think that shows that simply having more gameplay-oriented themes can inspire people to come up with designs of their own. Still, an outright "Original Puzzles" theme sounds fun as well.
Speaking of themes, there's still the cartoon character one to consider... perhaps I'm thinking too hard about this. Let's just say we'll do cartoon characters next, then original puzzles after that. Sound good?
@KoiTenchi: I haven't rushed the main game. I've been making so many levels (some you can't find anywhere, I keep them for me only) that I haven't had that much time to put into the main game. And 1/3 of the time the player is someone else than me as my friends really like playing it too and I want to see their reactions to my levels. But back to the question. And the answer is... 230! I've completed all the stages before it. Haven't tried 230 yet but I'm most likely going to beat it today.
Hey, isn't the contest over? I think that the winner's pushmo should be featured in the next contest, so that those who missed it can use the QR code to play it. EDIT-Oh, sorry, I didn't know what time it was, since I'm not in GMT.
Looks like we have a three-way tie! That's kind of awkward, but I guess I can just say that Ecto-1, Late and Portisanta are all winners! If you want all the glory for yourself, you'll just have to try again. I do hope this doesn't become a common occurrence, though.

I myself have never watched even a one episode as ponies really aren't my thing... But it looks really great. Don't yet know what I'm going to do this week. Haven't been into cartoons for some time.
