When you come across the first moving block, there's a room on the right containing the second gem.

Get to the room with all of the moving blocks and smash the bricks in the right hand corner to reveal the third gem.

Blizzard On The Star Express - Extra Challenge: Get The 1-Up Mushroom

Enter the engine room and tap (or press ZR) on the lever to release the 1-Up mushroom.
