I'm thinking they'll wait 2 months inbetween Directs, so I think August.
TBH, I'm hoping they go the route they've been going, and make the majority of upcoming Directs singular game focused (Like the Smash Bros. & Mario Kart ones), that way they won't be out of cards to play at E3, like what happened last year.
Ideally:
August - Hyrule Warriors Direct
October - Smash Bros Direct 2 (Focused on U version, since the first was 3DS focused)
December - Updated trailers of the 2015 games they highlighted at E3, with maybe one reveal
February - Single game Direct
April - Single game Direct
June - E3 event like this year's
etc, etc, etc...

Actually, I'd really love for them to do directs focusing on smaller stuff that's coming out before the next big even. Kinda like that February Direct. That said, they should also continue with single-game Directs. July might be a good time to talk about games coming to the eShop in August, September, and October. Maybe show off some more trailers. Hyrule Warriors playable character trailer. August could have a Hyrule Warriors Direct to get the hype for HW up, but there's no point in a September Direct. October, like you said, could be Smash U, but it could also focus on the myriad of games and eShop titles coming out in November. Lord knows, there's plenty of them. That said, it's all about what Nintendo feels we need to know and how often they think they have to tell us.
July - August seems to be a pretty good prediction for the next direct, but we'll see. Mostly depends on if Nintendo has any tricks up their sleeves when it comes to announcements or hyping upcoming games.
Anything not stated for release in the next two months I don't trust them on. (They think nothing of adding that much again but don't seem to do it when it is close to release.)
