Hands down DoReMi Fantasy. Quality graphics , sound (oh the sound!), controls and level design. Love it.

DoReMi Fantasy: Milon no DokiDoki Daibouken Not only is the much improved sequel to Milon's Secret Castle my favorite platformer on VC, but it's also my favorite VC download, favorite 16-bit game, favorite 2D platformer, and my number two favorite video game of all time A truly unforgettable experience, imo

I'm always incredibly jealous of everyone that seemed to have a much more enjoyable experience with DoReMi Fantasy than I did. I don't think I went into it with unreasonable expectations and I really wanted to like it, but I came away from it thinking that it was simply just a good, slightly more than basic platformer.
I'm not trying to "hate" on it or anything, I'm just genuinely envious of everyone that seemed to see something in it that I didn't.
