My most impressive feat was beating the main story of Star Ocean: Till The End of Time for the PS2 in under 12 hours during a speed run! I look forward to hearing other Nintendo Lifer's accomplishments!
I feel like I don't know is appropriate here. Theres been so many things that were such a challenge that I don't know what would be considered the greatest accomplishment.

Yesterday I beat the level 32 Shank on the original Castlevania using just the vampire killer and no II or III or anything like that.
I looked afterwards to see how other people did it on Youtube and everybody just used some spam of either the cross or holy water.
Castlevania 3 on the Wii VC without accepting any help (I didn't know what was going to happen and didn't want to start again so said no) on the Alucard route.
Best is probably the original Metal Slug MVS difficulty 1 credit clear.

I think it was getting the one billion point award in Pinball Fantasies: Billion Dollar Gameshow. I got so addicted to trying to get that jackpot I probably spent around 10 hours trying to get it.
Beating ActRaiser 2 on all difficulty settings
Almost beating the first Super Wagyan Land on the Super Famicom on the day that I got it (even though it was back before I used notes for the shiritori puzzles, which took me awhile to get to the end blind--a few hours at least)
Beating the final boss of Ys V Expert on my first try (considering the final boss in the previous four Ys games took me several tries to vanquish... not that it says much for Ys V's actual difficulty, really)
Beating Brain Lord in around seven to eight (non-consecutive) hours; if I could, I could try to beat it in less
There are so many great accomplishments that I've accomplished in retro titles, but I just cannot name them all
Me and a friend managed to play through the hovercraft stage in Battletoads together without losing a life - ironically, we lost nonetheless, at the rat race.
I beat Megaman 2 on 3DS VC without using save states(yeah I even took note of the "save" passwords and everything)
I'm not much of a retro gamer.... though I really wanted to play more retro games.

I beat AVGN vs Dr Wily
All Subspace Emissary Levels on Intense (That isn't retro, though)
Beat New Game + Shovel Knight
All without save states
I've never beaten him with the holy water, never even fought him with it. Always boomerangs or just the whip for me.
It is a bit like the cheap way that is in the video on Rondo of Blood to beat the 4 forms of Death with Maria. Get to kill him with holy water before whatever the things are he fires even get released.
The first one is quite reasonable I thought with just the whip it is the second quest one.
