Worst feeling while playing? Knowing that you are about to finish a high quality title that you don't want to end.
If you can't just replay the game and have just as much fun, maybe it wasn't such a high-quality title after all?
Once you know how the story ends, though... it's fun to replay, certainly, but you already know how it ends now. That same feeling of suspense is still there in the next playthrough, but it's never quite the same as the first time you ever beat the game, y'know?

No, if "not knowing how the story ends" is your main reason to keep playing rather than the game being entertaining on it's own terms, it's just an acceptable-quality game with a high-quality story.

One feeling I hate is that some SNES cartridges get dirty contacts and if you pop them into the SNES and they don't pull up the first time, it will sometimes erase your save data. So if I stop an RPG or something for awhile and then come back to finish it up, I always make sure to clean the contacts thoroughly before popping it into the console.
The worst feeling? Perhaps the absolute rage I felt during the final fight of the story in naruto clash of ninja revolution 3. That fight is so stacked it is insane.

A dull numbness and occasional sharp pains extending from my hand to elbow. In other words, the occasional resurgence of carpal tunnel due to not being careful enough to take it slow at work.

That fleeting moment right after you realize you saved over the wrong file. I remember putting 14 hours into Final Fantasy Mystic Quest (my first SNES RPG--got it as a present for good grades back in the day) and watching my brother save over that file literally the first time he touched the game. Especially as it's prompting "Do you want to save over this file?" and he selects "Yes" as I start to protest. And yes, he's almost 10 years older than me. Again, 14 hours isn't bad, but when you're in 3rd grade and that's one of your first end-of-game files.....not pleasant.

No, if "not knowing how the story ends" is your main reason to keep playing rather than the game being entertaining on it's own terms, it's just an acceptable-quality game with a high-quality story.
No, if you know the ending you can still enjoy the game, but you wont get the same feeling as the first time around, same thing as for books and movies. Now if the game have multiple endings on the other hand.
