That is an amazingly hard question. If I had to choose one game I'd go with Cave Story (and yes, I'm using that same "PC is current-gen" excuse, even though I've never played the PC version lol ), but a lot games are tremendously close seconds, and thirds, and fourths, and your can probably guess what's next, and so on. In a certain order:

I tore my hair out over this list, which is kind of sad. I rigorously chainsawed-down my Wii and DS lists to five each, while choosing my favorite game on the current-gen systems that I don't own or rarely play. I really just included PC so I could fit World of Goo in there and have a perfect list of 15 games. This is my second-favorite generation, just behind the GameCube one.
I'm going with Uncharted : Drake's Fortune/Among Thieves. I'm currently replaying them and still can't get over how beautiful they look in HD. They're insanely fun and the music is amazing! If I had to choose just one it would be Among Thieves but I love them both so much!

It interesting to me that all the best Wii games came early on it's cycle and 2 of them are Gamecube ports, whereas I think Sony's console is just hitting it's stride.
