Chuck E. Cheese for the DSiware. Greatest game evar. It will be remembered for generations.
But srsly, the Ocarina of Time, Majora's Mask, Gold/Silver, Earthbound, Mother 3, Melee, Kid Icarus Uprising, and Pikmin are all too good for me to choose just one.

BACK MY PROJECT EVEN $1 HELPS!
The best game ever is actually a random unfunny reference to a review series I haven't watched. Hey, I wasn't gonna let someone else say it first. (It's only okay when I do it while calling out myself and anyone else who would dare commit such a crime.)
I really don't know/can't decide. Chrono trigger is incredible (haven't finished yet), and I love Mario & Luigi Superstar Saga and Fire Emblem Sacred Stones is my favorite of the series. Portal 2 was literally flawless. Majora's mask was the best of the zelda franchise. Not much else comes close, besides maybe FE: Awakening and Bravely Default. And how could I forget Super Metroid and Fusion and Prime Trilogy. All incredible, but I can't say any one beats all the others.
Don't know about the best but the most time I've ever invested in a game was Diablo2, followed by Guitar Hero ( no haters please lol )
If we're going by time invested, then mine would easily be Neverwinter Nights. Not due to the game itself, but rather the tool set and what the player base did with it. So many persistent, player-driven RP worlds, and the most phenomenal RP experience I've ever had, which lasted for many, many years. Off the top of my head, I played the game heavily (on a daily basis) on several different worlds from 2002 to about 2008.
In terms of hours; Team Fortress 2. Ive invested more in that than in the entire Final Fantasy and Dragon Quest series combined (I play all the localised ones)
In terms of quality- I have a handful of masterpieces. Portal 2, Super Mario Galaxy, The Legend of Zelda: Wind Waker, The Legend of Zelda: Majora's Mask, Dark Souls, Papers Please, Shadow of the Colossus, Metal Gear Solid 3: Snake Eater, Half Life 2, Yoshi's Island.
AND FOR THE FAVOURITE!
SUPER METROID
This game is easily the most pure and refined game design I have ever played, with style and substance balanced like a yin-yang. Everything is tight, there is no fat. The visuals and soundtrack age with grace, like a fine wine. This is pure mastery to me.
Seriously, mine would probably be.....
Legend of Zelda: Wind Waker, Mass Effect 2, Mario 3, Batman: Arkham City, Red Dead Redemption, Shadow of the Colossus, Super Metroid, Pokemon Emerald, F-Zero GX, and Chrono Trigger (I don't even like these kinds of games, and I loved it).

Neverwinter Nights was awesome. I couldn't quite get a handle on all of the toolset but building my own campaigns was definitely a highlight in gaming for me. Even the campaign that shipped with it was very good. I'll never forget those environments.
