It's an internet prank where you trick someone into watching Rick Astley's "Never Going to Give You Up". If you get tricked, you've been "Rick Rolled". For an example, watch this video of a recent Shuttle launch.
As for the best Rick Roll ever? That would have to be Ben Heckendorn programming a Commodore PET to print out the lyrics. Totally epic experience.
