2 analog sticks, better battery life, and a more ergonomic design.

I never understood why Gameboys/DSs were always rectangular or square.
I don't have metal robot hands. My hands have curved surfaces.

dual analogs clickable, analog shift buttons and for nintendo to come to their sanity regarding eshop pricing, as for 3D I'd prefer they kept it but wouldn't be too bothered if they ditched it really.
