Besides Champion's Road, the Crown (hidden) level in Super Mario 3D Land. I could only beat it because of the Tanooki turning into a statue during the fuzzy part.
Perfect Run is hard only because you are not allowed to make a single mistake.

That one in the star world in NSMBU comes to mind where you have to hit the P switch and run as fast as you can to the next one before the time runs out.
I think it was called Run For It....that was tough. I must have died at least 40 times before I was able to get all 3 coins.
So I just finished my second playthrough of 3D World to get all the stamps and file stars. Used Mario this time, Toad was my first playthrough. Got to the dreaded Champion's Road.....
And beat it first try.
It's still a hard level, but once you get a feel for it, you know what to expect and how to handle it.

The special world from 3D land which has a crown in it is challenging if you don't use the tanooki suit. Perfect Run from Galaxy 2 is the challenging of all the levels in Galaxy 2.
Basically all Super Mario games have a fair challenging point that it just doesn't matter how hard one particular level is because if it is hard, just use a power-up that works for that level and that's just about it.
The only reason Lost Levels is hard is because it's a collection of tough levels with a strict life count. But the levels themselves aren't too tough. It's just that losing a life is more devastating. That being said, World 8-4 is a nightmare. Talk about pixel perfect platforming.
Lost Levels: 8-4, that's undoubtedly the most difficult one in my opinion!
