Sylux from Metroid Prime Hunters is my favorite. At times, I enjoy playing as Sylux more so than I've enjoyed playing as Samus throughout most of the Metroid Series. The thrill with Sylux is that Lockjaw form and how great it is in aerial travel. That travel is about as close as the 3D Metroids have ever gotten to transferring the infinite bomb jump over from some of the 2D games. Character-wise, the mystery of the hunter's identity, the hatred towards the Federation and the talents as a thief to steal from the Federation combined make for a background that is very interesting and easy to start making theories on. I really hope Sylux appears again in another Metroid game, the next if possible, or just give Sylux its own game.

This is kinda of a toughie considering such one-hit characters aren't generally memorable.
1. Ado from Kirby's Dream Land 3- No, he was not Adeleine from Kirby 64! He is basically a boy version of her, and he has more of an attitude compared to Adeleine's kindness. I know it's hard to tell, but I have a theory that they are twin siblings. He was an interesting and creative boss in Kirby's Dreamland 3. It makes me wish there were human or human-like characters in Kirby games again.
2. Ribbon from Kirby 64- Just a cute, little fairy, really. Again, I wish there were human or human-like characters in more Kirby games.

In addition to Adeleine & Ribbon from Kirby 64 and the Massif Bros. from Mario & Luigi: Dream Team; one of my favourite one timers is Luigi's Mansion's Melody.
That particular ghost really sticks out to me.
