Unless they come up with some crazy-awesome new IP. Mario & Luigi RPG 4 would steal the show for me.
That's because Superstar Saga, Partners in Time and Bowser's Inside Story are 3 games that are as close to perfect as it's possible for a game to get. If I was only allowed to play one series of games for the rest of my life, it'd be Mario & Luigi.
I've got a feeling that the September 13th event will have something to do with Kid Icarus Uprising and perhaps some discussion of the 25th Legend of Zelda Anniversary.
Seeing as Chocobo Dungeon 3D would be the business of Square Enix it is my hope that the 2011 Tokyo Game Show will serve as the premiere ground for an announcement of that along with a much wanted news of Kingdom Hearts: Dream Drop Distance, Final Fantasy Chocobo Racing 3DS and JUST maybe they'll reveal Batman Arkham for both the Nintendo 3DS and Sony PSVita. BTW Batman Arkham is based on the DC Comics Animation movie Batman Beyond: Return of the Joker.
It has to be Mother 4. That would surely be the best announcement I've heard in a long ass time.. Well, beisdes Smash Bros 4

But really, an extremely big title? What exactly could it be? It has to be bigger than Zelda, and I think Smash Bros. is a pretty big game for Nintendo... Nintendo RPG?

Let's hope it's Mother 4 although I EXTREMELY doubt it. It's sad though. I've HEARD so much about the Mother (Earthbound) games and yet I've NEVER been able to play a SINGLE one of them. I've seen them and know about them and like what I've seen but I've never ever been able to play a single one of them. Pity.
As much as I'd love the announcement to concern some big first party title(s), I bet it's something as mundane as Nintendo announcing an exact price of that new stripped-down Wii model.
I thought they might do an E3 and announce Four Swords DSiWare is ready to download as a part of it, like they did with LA. I think it'll be a showcase of software with one or two new titles shown off.

The media is speculating that Nintendo will either rename the 3DS (focusing less on 3D) or release a completely new handheld.
If the 3DS is getting discontinued, every single game that is in development will have to be cancelled.
If Nintendo is releasing a different handheld, then software developers will have even more work to do, and surprise announcements rarely work well (see the Sega Saturn).
If Nintendo ditches the 3D features, then all their games will have to be patched.
Nintendo isn't this stupid, are they? Then again there was the Virtual Boy... shudders
